This is a difficult question. Is this asking for the best performance as a villain in a superhero movie or best actor who happened to be a villain in a superhero movie? Until I get further clarification, I shall go back and forth between Heath Ledger and Jack Nicholson.

He was made for that Joker role, he acted The Joker perfectly, no, he WAS The Joker.
